    An Experiment with 3 Layers Development for IoT with NodeMCU12e + Nextion

    This paper presents the results of an experimental research with the development of a 3 layer solution for the Internet of Things using the NodeMCU12e controller and the Nextion touchscreen display to provide monitoring projects for individuals who require remote monitoring and special care, involving the health area, monitoring of patients, children or people who are in medical treatment who need a remote monitoring. The results demonstrated the possibilities for creating solutions that could serve as a basis for new projects with greater amplitude, contributing to more robust solutions that can meet different types of products that can collaborate with the health of the population in general

    In vitro antimicrobial sensitivity to Staphylococcus spp. isolates from dairy cows with subclinical mastitis

    Objetivou-se com este estudo avaliar a sensibilidade antimicrobiana in vitro de 291 isolados de Staphylococcus spp. recuperados de amostras de leite de vacas com mastite subclínica, em 15 propriedades rurais localizadas na Região Metropolitana do Recife (A), Agreste (B) e Zona da Mata (C) do estado de Pernambuco. Dos 291 isolados, 170(58,4%) foram classificados como Staphylococcus coagulase negativa (SCN), 84(28,9%) como Staphylococcus aureus e 37(12,7%) como Staphylococcus coagulase positiva (SCP). Para o estudo do perfil de sensibilidade a antimicrobianos empregou-se a técnica de difusão em discos, foram avaliadas 16 drogas antimicrobianas utilizadas no tratamento das mastites. O antibiótico que apresentou melhor eficácia in vitro foi a associação entre neomicina + bacitracina + tetraciclina com percentuais de 98,4%, 99,3%, 89,7% para as regiões A, B e C, respectivamente. O antibiótico menos eficaz foi a ampicilina que apresentou 56,5% de resistência para as amostras da região A, 72,8% para a região B e 71,8% na região C. Os resultados obtidos mostram a necessidade da realização periódica de testes de sensibilidade in vitro, pois existem variações no perfil de sensibilidade e resistência que podem comprometer o tratamento do animal bem como os programas de controle da mastite bovina causada pelo Staphylococcus spp.The objective of the investigation was to evaluate the in vitro antimicrobial sensibility of 291 isolates of Staphylococcus spp., taken from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife (A), Agreste (B) and Zona da Mata (C) in the state of Pernambuco, Brazil. From the 291 isolates, 170 (58.4%) were identified as negative coagulase Staphylococcus (SCN), 84 (28.9%) as Staphylococcus aureus, and 37 (12.7%) as positive coagulase Staphylococcus (SCP). To study sensitivity to antimicrobials, the diffusion in disks method was used with 16 antimicrobial drugs commonly employed in the treatment of mastitis. The most efficient antibiotic in vitro was the combination of neomicine + bacitracine + tetracycline with percentages of 98.4%, 99.3%, and 89.7% for the A, B, and C regions, respectively. The least efficient was ampicillin, which was resistant to 56.5% of the isolates taken from region A, 72.8% from region B, and 71.8% from region C. These results indicate the need for periodic testing of sensitivity in vitro, as these variations can compromise the treatment of animals as well as control programs for bovine mastitis caused by Staphylococcus spp

    Development of a Novel Biosensor Using Cationic Antimicrobial Peptide and Nickel Phthalocyanine Ultrathin Films for Electrochemical Detection of Dopamine

    The antimicrobial peptide dermaseptin 01 (DS 01), from the skin secretion of Phyllomedusa hypochondrialis frogs, was immobilized in nanostructured layered films in conjunction with nickel tetrasulfonated phthalocyanines (NiTsPc), widely used in electronic devices, using layer-by-layer technique. The films were used as a biosensor to detect the presence of dopamine (DA), a neurotransmitter associated with diseases such as Alzheimer's and Parkinson's, with detection limits in the order of 10−6 mol L−1. The use of DS 01 in LbL film generated selectivity in the detection of DA despite the presence of ascorbic acid found in biological fluids. This work is the first to report that the antimicrobial peptide and NiTsPc LbL film exhibits electroanalytical activity to DA oxidation. The selectivity in the detection of DA is a fundamental aspect for the development of electrochemical sensors with potential applications in the biomedical and pharmaceutical industries

    Sala de Aula Invertida no Ensino de Química: Planejamento, Aplicação e Avaliação no Ensino Médio

    O presente artigo apresenta os resultados da aplicação do modelo de sala de aula invertida na disciplina de Química com alunos do 3° ano do ensino médio. Foram preparados vídeo-aulas, quizzes e foi utilizada uma ferramenta wiki como ambiente virtual de aprendizagem. Durante a aplicação da metodologia, o conteúdo de Radioatividade foi abordado e os momentos presenciais foram utilizados para realização de resolução de questões e discussões. Questionários e um quizz foram aplicados com objetivo de avaliar a satisfação dos alunos em relação à metodologia vivenciada. Foi observado que a maioria dos alunos aprovou o método de sala de aula invertida, assim como o ambiente virtual de aprendizagem utilizado. A argumentação crítica, como também o raciocínio lógico mais aguçado e a superação das dificuldades relativas ao conteúdo foram observados e constituem evidências da contribuição do modelo de sala de aula invertida para a aprendizagem e desenvolvimento da autonomia dos alunos

    This study aims to understand the universalization of electricity consumption by poor communities in Brazil and raise the legal aspects and the universalization of electricity sector. The methodological procedures were based on qualitative strategy and documents analisys of two cases. Was used indicators of disadvantaged communities and sustainable energy development. The results show a tendency to make the power sector more socially inclusive, particularly for services universalization.Este trabalho objetiva compreender a universalização do consumo de energia elétrica por comunidades carentes no Brasil, levantar os aspectos legais e a universalização do setor de energia elétrica. Os procedimentos metodológicos basearam-se em estratégia qualitativa de base documental de dois casos estudados. Foram utilizados indicadores de comunidades desfavorecidas e de desenvolvimento energético sustentável. Tem-se uma tendência de tornar o setor elétrico socialmente mais inclusivo, principalmente pela universalização dos serviços

    Interleukin-6 increases the expression and activity of insulin-degrading enzyme

    FAPESP - FUNDAÇÃO DE AMPARO À PESQUISA DO ESTADO DE SÃO PAULOImpairment of the insulin-degrading enzyme (IDE) is associated with obesity and type 2 diabetes mellitus (T2DM). Here, we used 4-mo-old male C57BL/6 interleukin-6 (IL-6) knockout mice (KO) to investigate the role of this cytokine on IDE expression and activity. IL-6 KO mice displayed lower insulin clearance in the liver and skeletal muscle, compared with wild type (WT), due to reduced IDE expression and activity. We also observed that after 3-h incubation, IL-6, 50 and 100 ng ml(-1), increased the expression of IDE in HEPG2 and C2C12 cells, respectively. In addition, during acute exercise, the inhibition of IL-6 prevented an increase in insulin clearance and IDE expression and activity, mainly in the skeletal muscle. Finally, IL-6 and IDE concentrations were significantly increased in plasma from humans, after an acute exercise, compared to pre-exercise values. Although the increase in plasma IDE activity was only marginal, a positive correlation between IL-6 and IDE activity, and between IL-6 and IDE protein expression, was observed. Our outcomes indicate a novel function of IL-6 on the insulin metabolism expanding the possibilities for new potential therapeutic strategies, focused on insulin degradation, for the treatment and/or prevention of diseases related to hyperinsulinemia, such as obesity and T2DM.Impairment of the insulin-degrading enzyme (IDE) is associated with obesity and type 2 diabetes mellitus (T2DM).     Intragenic antimicrobial peptides (IAPs) from human proteins with potent antimicrobial and anti-inflammatory activity

    Following the treads of our previous works on the unveiling of bioactive peptides encrypted in plant proteins from diverse species, the present manuscript reports the occurrence of four proof-of-concept intragenic antimicrobial peptides in human proteins, named Hs IAPs. These IAPs were prospected using the software Kamal, synthesized by solid phase chemistry, and had their interactions with model phospholipid vesicles investigated by differential scanning calorimetry and circular dichroism. Their antimicrobial activity against bacteria, yeasts and filamentous fungi was determined, along with their cytotoxicity towards erythrocytes. Our data demonstrates that Hs IAPs are capable to bind model membranes while attaining α-helical structure, and to inhibit the growth of microorganisms at concentrations as low as 1μM. Hs02, a novel sixteen residue long internal peptide (KWAVRIIRKFIKGFISNH2) derived from the unconventional myosin 1h protein, was further investigated in its capacity to inhibit lipopolysaccharide-induced release of TNF-α in murine macrophages. Hs02 presented potent anti-inflammatory activity, inhibiting the release of TNF-α in LPSprimed cells at the lowest assayed concentration, 0.1 μM. A three-dimensional solution structure of Hs02 bound to DPC micelles was determined by Nuclear Magnetic Resonance. Our work exemplifies how the human genome can be mined for molecules with biotechnological potential in human health and demonstrates that IAPs are actual alternatives to antimicrobial peptides as pharmaceutical agents or in their many other putative applications
